CrO©ý-oxidated, de-esterified, and carboxyl-reduced products of PA-¥±a-1 were methylated to investigate the anomerization and glycosidic linkage composition. In native PA-¥±a-1, 2-linked Xylf, 5-linked Xylf, and 2-linked Fucf residues existed as methyl esters. Most ¥á-mannosyl and ¥â-glucuronosyl residues were observed as pyranoses. Most xylosyl and fucosyl residues had ¥â-configuration, whereas galactosyl residues had both a-and ¥â-configurations. CS-1, a periodate-resistant back bone, was obtained from the periodate oxidation and controlled Smith degradation of PA-¥±a-1. It was either hexa- or heptasac-charide and was composed of D-mannose and D-glucuronic acid in the molar ratio of 5.65 : 1.57. By combining results from ©öH- and ^(13)C-NMR spectroscopy, methylation analyses using GLC and GC/MS, and two dimensional ©öH-©öH COSY analysis, the structure of CS-1 was deduced as a ¥â-D-glucurono-¥á-D-(1¡æ2)-linked mannan.
